Absorption
Drug absorption is the process of movement of unchanged
drug from the site of administration to the systemic
circulation.
Except when given i.v., the drug has to cross biological
membranes by any of the mechanism of drug transportation
as described earlier.
A drug given as aqueous solution is absorbed faster than
when the same is given in solid form or as oily solution.
Increased blood flow increases drug absorption.

Oral Route
Absorption from stomach is slower as compared to small
intestine because in the stomach mucosa is thick, covered
with mucus and the surface area is small while absorbing
surface area of small intestine is much larger due to villi.
Presence of food dilutes the drug and retards absorption.
Most drugs are absorbed better if taken in empty stomach.
Certain drugs form poorly absorbed complexes with food
constituents, e.g. tetracyclines with calcium present in milk.

Gastro resistant tablets and sustained release preparations
can be used to overcome acid lability, gastric irritancy and
short duration of action.

The oral absorption of certain drugs such as digoxin and
cyclosporine is low because a fraction of the absorbed drug
is expelled back into the intestinal lumen by the efflux
transporter P-gp (P-glycoprotein) located in the gut
epithelium.
