Enzyme evolution and the temperature dependence of enzyme catalysis
The article discusses how enzymes are temperature dependent and how they have
evolved and function under certain conditions. For example, enzymes from organisms living in
cold environments have different optimal temperatures than those living in hot environments.
They compare thermophilic enzymes (more rigid) to psychrophilic enzymes (less rigid). They
mention a concept called enthalpy-entropy trade off (enzymes reduce the activation energy for
reactions at different temperatures by making a trade off between enthalpy and entropy. They
gave an example on how thermophilic (heat loving enzymes) are less active at lower
temperatures due to the tradeoff. They talk about Nonlinear Arrhenius behavior (didn’t really
understand this part). Lastly, they talk about the role that temperature plays in enzyme activity
and how it can be studied through experimental techniques such as x-ray crystallography,
computational simulations, and Cryogenic electron microscopy.

Enzyme Therapy: Current Challenges and Future Perspectives
The article highlights the diverse roles of enzymes in therapeutic applications across
several pathologies. They talk about several therapeutic treatments with enzymes that have been
put into place to treat and manage the symptoms of the following disorders. Lysosomal Storage
diseases (LDL): enzyme replacement therapy has proven effective in treating this rare group of
genetic disorders caused by enzyme deficiencies that lead to an accumulation of specific
molecules within the cell. Other disorders include PKU (phenylketonuria) (rare inherited
disorder that causes the amino acid phenylalanine to build up in the body. This is significant
because it can result in brain damage). Those are just some of the diseases/ disorders that the
article talks about. Lastly, it also mentions how the use of enzymes in therapies have an evolving
and seem promising in the field of biotechnology with a huge potential to offer new treatment
options for a wide range of pathologies. Researchers still must do extensive research and address
the challenges associated with the treatments including the immune responses of patients to
make sure that the therapies are safe and effective.
