ACTIVITY NO. 3 - ENZYME ACTIVITY

Introduction: Enzymes are biological products produced by our body to help speed up chemical reactions, for
instance, food digestion. They break down other carbohydrates, proteins, and fats. Enzymes catalyze such
chemical reactions by reacting with a substrate. In this experiment, let’s take a look at the reaction of an
enzyme, its substrate and their product.

Materials:
- Safety goggles or protective glasses - Tap water
- Liquid dish soap (5 teaspoons) - Measuring cup
- Dry yeast, 1 packet (7 grams) - Permanent marker
- Tablespoons (kutsara) - Food coloring (yellow, blue, red, and green)
- Teaspoon (kutsarita)
- 5 Plastic cups, 16 oz
- Hydrogen Peroxide, 3% (Agua Oxigenada, at least 100 mL)

Instructions:
A. Prep Work
1. In a plastic cup, dissolve 1 packet of yeast in a ½ cup warm water (DO NOT use hot water).
2. Use a teaspoon to mix the solution. Let the dissolved yeast rest for at least five (5) minutes.
3. Set aside.

B. Experiment Proper
1. Using the permanent marker, label the remaining 4 plastic cups with numbers 1, 2, 3, and 4.
2. Add 1 teaspoon of liquid dish soap in each labeled cup.
REMINDER: Before using the hydrogen peroxide, put on your safety goggles to protect your eyes. In case you spill
hydrogen peroxide, clean it up with a wet paper towel. If you get H₂O₂ (hydrogen peroxide) on your skin, make sure to
rinse the affected area with plenty of water.
3. Put on your safety goggles.
4. To cup 1, no further addition is necessary.
5. Using a clean spoon, add ½ tablespoon of 3% hydrogen peroxide solution to cup 2.
6. Add 1 tablespoon of 3% hydrogen peroxide solution to cup 3.
7. Add 2 tablespoons of 3% hydrogen peroxide solution to cup 4.
8. To each labeled cup, add the following food coloring: red for cup 1, blue for cup 2, yellow for cup 3 and green for cup
4.
9. Using a clean tablespoon, add 1 tablespoon of the dissolved yeast solution to cup 1and swirl the cup lightly to mix the
solution. Observe what happens after the addition of the yeast.
10. Add 1 tablespoon of the dissolved yeast solution to cup 2 and swirl the cup lightly to mix the solution. Observe what
happens after the addition of the yeast. Observe what happens after the addition of the yeast.
11. Add 1 tablespoon of the dissolved yeast solution to cup 3 and swirl the cup lightly to mix the solution. Observe what
happens after the addition of the yeast. Observe what happens after the addition of the yeast.
12. Add 1 tablespoon of the dissolved yeast solution to cup 3 and swirl the cup lightly to mix the solution. Observe what
happens after the addition of the yeast. Observe what happens after the addition of the yeast.
13. Place all 4 cups next to each other in front of you and observe your results.
14. Take cup 1 and add 1 additional tablespoon of the 3% of the hydrogen peroxide solution.
