1 Operating instructions
1.1 Using this manual

 Make sure that the service manual and the operating manual are available in the latest versions. To do
so, compare the version numbers. The latest versions of the manuals can be downloaded from
www.eppendorf-support.com.
 Read the service manual before commencing work on the device.
 Read the chapters "Installation" and "Operation" of the operating manual.
 Observe the safety instructions in the operating manual.

1.4 Purpose and scope


1.4.1 Purpose

This document describes all prerequisites and actions necessary to perform installation, diagnosis, repair
or maintenance of the product named on the title page.

1.4.2 Scope

This document applies to all service organizations of Eppendorf AG, service providers commissioned by
Eppendorf AG, as well as certified Eppendorf AG service technicians.

2.2 Features

The Reference 2 pipette is a piston-stroke pipette for aspirating and dispensing liquids. The pipette
operates according to the air cushion principle. A suitable pipette tip must be fitted prior to use. The
dispensing, volume setting ring and ejector are controlled with the control button. Depending on the
model, volumes between 0.1 μL and 10 mL can be dispensed.

2.2.1 Pipette models

Different variants are available:


• Single-channel pipettes with fixed volume (fix)
• Single-channel pipettes with variable volume setting
• Multi-channel pipettes with 8 or 12 channels and variable volume setting

3 Safety
3.1 User profile

The specialist entrusted with maintenance, repair or testing must meet the following prerequisites:
• Successful participation in service trainings with certification by Eppendorf AG for the product that is to
be maintained, repaired or tested.
• Qualification as a specialist with knowledge of the applicable local and international standards.
• Qualification to evaluate the work assigned to him.
• Recognition of potential hazards and how to prevent or correct them.

3.2 Information on liability

The Eppendorf service organization responsible for servicing, repair, or testing is liable for ensuring that all
work is performed in a professional manner.

 The constructive condition of the device must not be altered as a result of repair, servicing, or testing.
The safety of the original condition must be maintained.
 Only accessories and original spare parts recommended by Eppendorf as well as test and measuring
equipment recommended by Eppendorf may be used for servicing, repair, and testing.

WARNING! Damage to health due to infectious liquids and pathogenic germs.

 When handling infectious liquids and pathogenic germs, observe the national regulations,
the biosafety level of your laboratory, and the manufacturers' Safety Data Sheets and
application notes.
 Wear your personal protective equipment.
 For comprehensive regulations about handling germs or biological material of risk group II
or higher, please refer to the "Laboratory Biosafety Manual" (source: World Health
Organization, Laboratory Biosafety Manual, in the currently valid version).

WARNING! Damage to health due to toxic, radioactive or aggressive chemicals.

 Wear your personal protective equipment.


 Observe the national regulations for handling these substances.
 Observe the material safety data sheets and manufacturer's application notes.

There may be contaminated material on the device and the accessories. You may become
infected by contaminated material.

 Find out more about the risk of contamination before starting work.
 Check the device decontamination certificate of the device.
 Work on decontaminated devices only.
 Wear your personal protective equipment (protective gloves, protective goggles).

impair the safety, functioning and precision of the device. Eppendorf cannot be held liable or
accept any liability for damage resulting from the use of accessories and spare parts other
than those recommended or from the improper use of such equipment.

 Only use accessories and original spare parts recommended by Eppendorf.

4 Operation
4.1 Setting the volume

Set the volume from a high to a low value. If necessary, exceed the desired volume and then
go back.
1. Keep the volume catch pressed.
2. Turn the control button.

4.2 Attaching the pipette tips

The pipette tip can be attached by hand or mounted directly from a tip holder (tray) with the pipette.

The pipette tips are single-use items.

The control button and the trays are color-coded. The color marks the pipette size and the
volume of the pipette tip (epT.I.P.S.).
1. Insert the tip cone into the pipette tip using light
force.

4.3 Optimum immersion depths


Volume Immersion depth
0.1 μL – 1 μL 1 mm
1 μL – 100 μL 2 mm – 3 mm
100 μL – 1000 μL 2 mm – 4 mm
1 mL – 10 mL 3 mm – 5 mm

4.4 Forward pipetting


4.4.1 Aspirating liquid

Prerequisites
• The pipette tip has been attached.

To ensure maximum precision and accuracy, we recommend initially wetting each new tip by
aspirating and dispensing the liquid one to three times.

1. Press down the control button to the first stop.


2. Immerse the pipette tip vertically in the liquid.
3. Maintain the immersion depth and let the control button slide back slowly.
Liquid is aspirated into the pipette tip.
4. Wait until the liquid has been aspirated.
5. Remove the pipette tip from the liquid.

If necessary, wipe the pipette tip against the tube inner wall.

4.4.2 Dispensing liquid


1. Place the pipette tip on the tube inner wall at a steep angle.
2. Press down the control button to the first stop.
The liquid is dispensed.
3. Wait until the flow of liquid stops.
4. Press down the control button to the second stop.
The pipette tip is completely emptied.
5. Hold down the control button and wipe the pipette tip on the tube inner wall.

4.4.3 Ejecting the pipette tip

 Push the operating lever all the way down.


The pipette tip is ejected.

4.5 Reverse pipetting

An additional volume (blow out) is aspirated when performing reverse pipetting. This can improve
dispensing results for viscous or foaming liquids. Volume limits may occur when using filter tips.

4.5.1 Aspirating liquid

1. Press down the control button to the second stop.


2. Immerse the pipette tip vertically in the liquid.
3. Maintain the immersion depth and let the control button slide back slowly.
Liquid is aspirated into the pipette tip.
4. Wait approx. 3 seconds.
5. Remove the pipette tip from the liquid.

If necessary, wipe the pipette tip against the tube inner wall.

4.5.2 Dispensing liquid

1. Place the pipette tip on the tube inner wall at a steep angle.

6.2 Assembling the single-channel pipette lower part up to 1000 μL

Please complete the following steps in the order described:

1. Carefully guide the piston and the piston spring into the cylinder.
1 Make sure that the piston is guided correctly in the piston spring
and the cylinder. There must not be any perceptible resistance.
Stop pushing as soon as any resistance is felt. This may indicate

that the piston is not positioned correctly in the cylinder. There is
a risk of bending the piston if too much force is applied. Carefully
pull out the piston and repeat the process correctly. In the case of
piston springs with double windings (4), these windings must
3 point downwards.
2. Keep the piston and the piston spring pressed.
3. Keep the stop pins on the piston mount pressed with the other
4 hand.
4. Mount the piston mount so that the two locking mechanisms
engage in their holders.

5. Gently press a pipette tip against the inserted piston. The piston
must move down in the cylinder without any noticeable
resistance.
6. Insert the pre-assembled lower part of the pipette into the upper
part until it audibly engages.
7. Place the ejector sleeve on the ejector rod. If correctly fitted, it
will engage slightly.

7 Alignment/adjustment
7.1 Adjusting the pipette
NOTICE! Incorrect dispensing volume with special liquids and due to temperature
differences.
Solutions with physical data very different to that of water or temperature differences between
the pipette, pipette tip and liquid may result in incorrect dispensing volumes.

 Avoid temperature differences between the pipette, pipette tip and liquid.

The random and systematic errors recorded upon delivery can be found on the Eppendorf
Certificate. This certificate is included in the delivery. If the factory adjustment is changed, the
certificate becomes invalid.

7.1.1 General information on user adjustment and the factory adjustment

The Reference 2 was adjusted and checked before delivery. The adjustment opening is sealed with a gray
plastic adjustment seal. The adjustment seal bears the abbreviation "ADJ". The adjustment display shows
"0".

Changing the adjustment of the Reference 2 is recommended for solutions whose density, viscosity, surface
tension and/or vapor pressure differs greatly from that of water. If the density of an aqueous solution
changes by approx. ±10 %, for example due to its salt concentration, its volume changes by approx.
±10 %. This does not apply if other relevant properties of the liquid also change.

If the location the pipette is used at is at an extreme altitude, correction due to atmospheric pressure is
required. At 1 000 m above sea level, a volume error of approx. -0.3 % occurs for a 100 μL pipette.

If special tips with shapes different from standard tips are used, changing the adjustment can improve
dispensing accuracy (systematic bias).

Observe the recommendations for adjusting epT.I.P.S. long in the User adjustment on the Reference 2 CD.

A changed adjustment can be cancelled in a few simple steps.

Aside from changes to the user adjustment, a Reference 2 can be changed permanently by changing the
factory adjustment.

Changes to the user adjustment or to the factory adjustment have no influence on dispensing precision
(random error). Precision can be improved by replacing worn parts. Precision is also greatly influenced by
the handling of the device.

Before changing the adjustment of factory calibration, check the existing dispensing rate.

You can determine the actual volume by weighing:

At 20 °C, the density of distilled water is approx. 0.9982 mg/μL, and 0.9965 mg/μL at 27 °C.

No correction is required if the set volume equals the actual volume.

If there is a difference between the actual volume and the set volume of distilled water, please check the
following:

• Is liquid dripping from the tip?


• Is the pipette tip leaking?
• Is the tip cone damaged?
• Are the piston and cylinder leaking?
Sufficient tightness is ensured if no drops from at the pipette tip after aspirating the nominal volume of
distilled water and a wait time of approx. 15 s. Hold the pipette vertically and do not touch the pipette
tip during this. For nominal volumes of ≤ 20 μL, pre-wet the tip multiple times.
• Is the temperature of the pipetted liquid the same as the:
– Temperature of the device?
– Temperature of the ambient air?
• Is the measuring place draft-free?
• Does the operation and the pipetting speed ensure complete aspiration and dispensing of the liquid?
• Has the correct numerical value for "Dense liquid at weighing temperature" been used to calculate the
actual volume?
• Is the set volume correct?
• For very small volumes (< 10 μL): Is the analytical balance sensitive enough (Scale resolution:
0.001 mg)?
• Have original epT.I.P.S pipette tips been used as test tips?
The pipette tip used as a test tip can be found in the technical data (see p. 47).

Only if all questions can be answered with "Yes" can adjustment be performed. In any other case, the
problems corresponding to the questions answered with "No" must first be remedied. If replacing the
complete lower part or other volume-determining parts, proper assembly must be gravimetrically checked.
The systematic and random errors to be fulfilled can be found in the technical data.

7.1.2 Changing the user adjustment

If the adjustment is changed, the volume is changed by a specific value. Strictly speaking, this change only
applies for the testing volume.

Example:

You are adjusting a 10 – 100 μL pipette by 1 μL at a volume setting of 100 μL (1 μL ≙ 1 %). The pipette is
also adjusted by 1 μL at a volume setting of 10 μL (≙ 10 %).

For instructions on how to disassemble and assemble the pipette, read the corresponding
chapter "Disassembly/assembly".

Gravimetrically check the pipette if volume-determining parts have been replaced.

9.1.1 Single-channel pipettes

1. Check if the lower part is contaminated. Replace contaminated lower parts.


2. Check if the inserted lower part is bent. Replace bent lower parts.
3. Clean the tip cone using an alcohol-based solution. Inspect the tip seating for scratch marks. Replace
scratched lower parts.
4. Use the pipette tester to check if the pipette is leaky. Replace the piston with seal if necessary, then
repeat the test.
5. Check if the piston is broken or scratched. Replace broken or scratched pistons.
6. Clean the piston using an alcohol-based solution, then lubricate the piston.
7. Assemble the pipette. Set the nominal volume. Check to see if the piston runs smoothly and evenly by
slowly pressing the control button several times.

9.1.2 Multi-channel pipettes

1. Remove the tip cone O-rings using the multi-channel tool.

If the O-rings cannot be cut, the tool is worn and must be replaced.

2. Check if the tip cones are scratched. Replace scratched tip cones.
3. Place the new O-rings on the tip cones.
4. Check the piston along with the seal. Replace defective pistons with seals.
5. Clean the piston using an alcohol-based solution, then lubricate the piston.
6. Use the pipette tester to check each channel for leakage.
7. Assemble the pipette. Set the nominal volume. Check to see if the piston runs smoothly and evenly by
slowly pressing the control button several times.

10.7 Test conditions

Test conditions and test evaluation in compliance with ISO 8655, Part 6. Tested using a standardized
analytical balance with evaporation protection.

The three largest testing volumes per tip (10 %, 50 %, 100 % of the nominal volume)
correspond to the specifications in accordance with ISO 8655, Part 2 or Part 5. The test is to
be carried out with these three testing volumes for testing of the systematic and random error
in compliance with the standard. The smallest adjustable volume serves to provide additional
information.

• Water according to ISO 3696
• Inspection at 20 °C – 27 °C
