Discussion (Aina)

In plant tissue culture, there are some major steps to be followed to get a better results

of cultured cells. The first phase is initiation phase where tissue culture is obtained and

sterilized in order to prevent any microorganisms from negatively affecting the process. Next

is the multiplication phase which is the second step of tissue culture where the in vitro plant

material is re-divided and then introduced into the medium. Here, the medium is composed of

appropriate components for growth including regulators and nutrients. These are responsible

for the proliferation of the tissue and the production of multiple shoots. The last phase is the

root formation where the roots are formed. At this phase, hormones are required in order to

induce rooting and consequently complete plantlets.


There are some precaution steps that should be aware while handling this experiment.

Firstly, wear an appropriate PPE such as gloves and lab coat to avoid contamination with the

tissue culture and also always kept the laboratory cleaned regularly. Besides that, we must

sterilize all laboratory equipment such as pipette, beaker, forceps and so on before use to

avoid contaminate substances. We also have to make sure that the cell culture is in a

sterilized environment. We should transfer the cell culture from the incubator to hoods

immediately because it cannot remain outside for a prolonged period as they could affect the

results of the cell culture. Last but not least, we must check the cultured cells regularly within

two weeks period to make sure that the cultured cells safe from being contaminated.

Conclusion

In a nutshell, there are numerous methods for plant tissue culture that provide certain

advantages such as the production of multiple plant that require specific conditions for the

culture process obtain desired results. On the other hand, plant tissue culture may be used for

cloning purposes, genetic modification of a given plant and also to increase yield of the plant

of interest. Therefore, tissue culture is a great significance in biological studies due to its

wide range of applications, this technique also is significant to many biological fields

especially botany, agriculture and forestry.
