Problem:
How is the microscope used?

Materials:
 Microscope  Scissors
 Lens paper  Newsprint
 Slides  Magazine
 Coverslips  Dropper
 Water  Prepared Slide

Procedure:
1. Cut a piece of newsprint that contains the letter “e”.
2. Place the letter “e” in the center of the slide.
3. Using a dropper, place a drop of water on the letter “e”.
4. Place a coverslip at about a 45 degree angle over the drop of water. Gently lower the
coverslip onto the slide. Tap the coverslip gently to remove air bubbles.
5. Place the wet mount slide of the letter “e: on the stage of the microscope with the “e”
facing you as you would read it. Adjust the “e” so that it is over the opening of the stage.
6. Turn the nosepiece so that it is on the lowest power objective (4x).
7. Raise the stage while looking at the slide from the side.
8. Now look into the eyepiece and use the coarse adjustment to bring the “e” into focus.
9. Use the fine adjustment to sharpen the focus.
10. Draw the letter “e” in the circle (a).
11. Turn the nosepiece so that the medium power objective (10x) is in place. Use the fine
adjustment only to focus!
12. Draw the letter “e” in circle (b).
13. Turn the nosepiece so that the high power objective (40x) is in place. Use the fine
adjustment only to focus!
14. Draw the letter “e” in circle (c).

Observations:
1. While looking through the microscope, in what direction does the “e” appear to move
when you move the slide?

2. Determine the magnification of your microscope (show your work).
a. Low power: eyepiece ____ x objective ____ = _____ mag.

3. Procedure: Colored Paper
a. From the magazine or funny paper cut out a small piece of colored paper. Prepare
this slide the same as the “letter e” slide. Use the 10x objective. How does this
compare with the picture as seen with the naked eye? Using your “Microscope
Drawing” Sheet make a sketch of the colored paper as it appears under the
microscope.

4. Label the parts of the microscope below

5. Your teacher has some “prepared” slides for you to observe. These are slides that are already
prepared for you to view. Using your “Microscope Drawing” Sheet to make a sketch of 5
“prepared slides” as they appear under the microscope. Be certain to label your sketches on
your “Microscope Drawing” Sheet with the proper Title, Magnification, and Description for
each slide observed.
