4.. Methods / Procedures


4.1 Overall Process of This Project
This research work will involve with several phase of work including solubility curve and
crystallization of fructose in different solvent. Different solvent have different solubility
curve. In the experiment, during the analysis procedure, micro siever is used for
distribution of particle size of crystal, Fructose crystal habit is determined under
microscope and texture analyzer is used to determine the crystal hardness are the
equipment used to analyze the result of the experimental procedure. Furthermore,
Fourier-Transformer Infrared (FTIR) is also used to confirm the crystal of fructose.

Preparation of Material

The raw materials had been used are Fructose, Ethanol,water (H 2O) Beaker,
Test tube, Measuring Cylinder, Thermometer, Magentic Stirrer, Spatula
and Filter Paper.

Preparation of Fructose Sample

A 100g of fructose sample is weighted and put in 200ml beaker. Fill the
beaker with 100ml of distilled water and mixed the solution on the hot plate
until the solution is dissolve at 20℃.
The solution was filtered with filter paper and the mixture is dried at 50℃
for 2 hours in the oven.
The crystal sample is placed into the dessicator and the amount of fructose
and water mixture is observed and weighted.
The step is repeated for other solvent of water + ethanol

Analysis of Product

The product obtain are analyse using:

Microscope
Texture Analyzer
Fourier-Transformer Infrared (FTIR)

4.2 Experimental Procedures
4.2.1 Preparation of Fructose Sample

4.2.1.1 Determination of Fructose Solubility in Different Solvent


The formation of the fructose crystal is depending on the solubility of fructose in solvent.
The solubility curve is important in determining the solubility of fructose because
different solvent have difeerent solubility.

4.2.1.1.1 Crystallization of Fructose in the Solvent


1. 100g of fructose sample is weighted and put in 200ml beaker
2. Fill the beaker with 100ml of distilled water and mixed the solution on the hot
plate until the solution is dissolve at 20℃
3. The solution was filtered with filter paper to remove the excess residue
4. The mixture is dried at 50℃ for 2 hours in the oven
5. The crystal sample is placed into the dessicator and the amount of fructose and
water mixture is observed and weighted
6. The step is repeated for other solvent of water + ethanol

4.3 Analysis Porcedures

All the samples and final products are analyzed to find out their characteristics and
properties.

4.3.1 Particle Size Distribution for Crystal by Using Micro Sive shaker
1. 100g of fructose is weighted and grinded about 3 minutes.
2. The grinded fructose is placed in the micro sieve shaker with suitable
amplitude setted.
3. The amount of samples in each sieve of respected size is being weighted
and recorded.

4.3.3 Determination of Fructose Crystal Habit by Using Microscope


1. The crystal grain of crystallized fructose with different solvent will be
placed under the microscope.
2. The knob of the micrscope will be adjusted until the image of the crystal
become clear.
3. The image of crystal shape and habit under the microscope will be
captured and examined.

4.3.3 Determination of Crystal Hardness by Using texture Analyzer


1. The Crystallized sample was sliced into equal size
2. Before carrying out the test, the texture analyzer will be calibrated to
acknowledge the texture analyzer test surface.
3. The probe was set lower which is closer to the test surface to calibrate the
equipment.
4. The ‘calibrate’ in the menu bar is clicked to specify thedistance return of
the probe which is about 30mm.
5. The “Run a test” step was run and the Auto eight box before commencing
the test (X) was also checked.
6. The fructose sample was placed centrally under the cylinder probe.
7. The compression test was commenced, the firmness was then recorded.

4.3.4 Fourier-Transform Infrared (FTIR) Spectroscopy Test to Confirm the Crystal
of Fructose
1. The FTIR is used to analyze the changes in the chemical structure of the sample.
2. The sample need to be grinded first into fine powder using pestle mortar before
testing it for FTIR.
3. The samples are analyzed and the absorbance is obtained.
4. The spectra of organic compounds for the fingerprint region is from 400-1500cm-1
and the functional group region is from 1500–4000cm-1.
5. The presence of bonded acid species on the solid acid catalyst can be proved by
the range of vibration band from 1500–4000cm-1.

and surrounding, comfortable and wide space of workplace, clean as well as sufficient oxygen to

8. Specific risk assessment for procedure and precautions to be
taken

During performing the experimental activity such as in the preparation of catalyst, there is a
LOW risk of contact with hazardous chemicals which is sulphuric acid (H2SO4) if all the
precautionary measures stated in Table 5a and 5c(handling) are followed well. Extra care must be
taken to avoid chemical spillage or other incident to happen, hence direct exposure to chemical
can be prevented. PPE must be worn at all time during conducting the experiment. Work in well-
ventilated area such as fume hood. Before starting any steps or process of procedures, ensure
that all apparatus and equipments must are in a good condition.

During sample analysis using the HPLC,FTIR and NMR, there is a LOW risk of contact with
chemical since the sample solution contains low concentration. Avoid aerosol and fume formation
by selecting a modertae strirring speed. Turn on the electrical appliance ONLY after all of the
component have been connected and the cables checked for any damage. To conduct the unusual
handle the equipment, please refer to the guideline book of the equipment or can be refer to
person in charge. Hence, the safety and precautions steps of overall risk assessment for this
procedure is LOW.
