AP Biology Pre Lab #2

This document provides the pre-lab assignment for an AP Biology lab on enzyme catalysis. The lab involves using the enzyme catalase to catalyze the decomposition of hydrogen peroxide and measuring the reaction rate over time. Students are asked to answer 21 questions to demonstrate their understanding of the purpose, variables, methods, and expected results of the lab experiment. This includes identifying the independent variable as time, the dependent variable as volume of hydrogen peroxide decomposed, and that the reaction rate is expected to decrease over time as the reaction progresses.

LAB #2: ENZYME CATALYSIS

LAB ASSIGNMENT
Name__________________________________ Date__________________________

PRE-LAB
Read Lab Two on pages 19-28 of your laboratory textbook, then answer the
following questions in complete sentences.
1. What is the function of catalase in cells? Write the equation for the reaction
catalyzed by catalase.

2. What are the purposes of the exercises in this lab?

Exercise

Purpose

2A
2B
2C
2D

3. In the lab procedure, what is the function of sulfuric acid?

4. How are you going to determine the amount (in mL) of hydrogen peroxide
remaining after the enzyme has been allowed to react for a specified time
period?

5. What is the difference between the procedure in the baseline assay (Part 2B)
and that of the experimental assays (Part 2D)?

6. Why is it important to continually swirl the contents of the beaker after the
catalase has been added to the hydrogen peroxide?

7. State a hypothesis for Exercises 2D.

8. Briefly explain how you reached your hypothesis. Describe the logic that you
used to reason from what you know about the learning context to your educated
guess of the outcomes of the experimental procedure.

9. Identify the independent variable in Exercise 2D

10. Identify the dependent variable (i.e. what is being measured) in Exercise 2D.

11. Identify any other variables that are being held constant in the experimental
group. For instance, each step may be, measured for the same amount of time;
or each setup may have the same amount of solution in each beaker, etc.

12. What method and/or time frame is used?

Example: I will take readings of..every 5 minutes for 30 minutes.

13. What is the rate of calculation and/or statistical application?

Example: Average number of trials, slope of the curve, etc.

14. How will the results be verified? (Sample size or repetition)

15. How will the experimental results be presented? (Graphs, charts, etc.)

16. What are the expected results for Exercise 2D? Why? This is your best
prediction based on reading the lab and any other information you obtain. (Other
sources on information can be your book, your notes, Cliffs book)

17. Given the following data that might be taken in Part 2D, complete the table.

18. On the graph below, plot the volume of H2O2 uses as a function of time, with title,
properly labeled axes, etc. Draw a curve as described in the introduction of the
lab.

19. Determine the initial rate of the reaction by finding the slope of the fitted line, with
units, for the interval 0-10 seconds. Show your work (with units). Give the
answer with correct units and significant figures.

20. In the same way, find the reaction rate for each of the time intervals; record them
in the table below.

21. Explain why the reaction rate changes over time.

22. Go to the online text book, click on Lab Bench. Complete the exercises and
animations for this lab, and take the Lab Quiz.
a. What was your score on the lab quiz?

b. Which questions did you miss?

c. What questions do you have about concepts/ideas you do not

understand?
