Application of OrganoGel in Metabolism and Toxicology Research The methods of drug toxicological research are primarily animal experiments, which extrapolate the effects observed in experimental animals to humans. The main aspects of new drug toxicity research include: acute toxicity studies, long-term toxicity studies, formulation toxicity studies, and special toxicity studies. The experimental methods are mainly divided into in vivo and in vitro tests. In vitro tests utilize isolated organs, cultured cells, cell organelles, and microorganisms for toxicity research. Traditional 2D cell cultures cannot construct a three-dimensional microenvironment, thus there is a significant difference in cell function compared to in vivo conditions. Three-dimensional (3D) culture uses special methods and materials to allow cells to grow in a three-dimensional manner, more closely resembling the growth patterns within the body, forming microstructures similar to those of tissues. 3D cell culture is mainly divided into scaffold-based 3D cell culture and scaffold-free 3D cell culture. Scaffold-free 3D cell culture is a simple and easy-to-operate method that does not require the addition of cell scaffolds. Instead, it utilizes cell culture vessels treated with cell-repellent materials or employs special systems such as hanging drop or microfluidic devices for cell cultivation. This prevents cell adhesion to the vessel surface, forcing them to aggregate and form cell spheroids. The 3D cell models obtained through this method possess a complete spherical structure, can spontaneously form extracellular matrix, and effectively simulate the oxygen and nutrient gradients found in in vivo environments.

Location: School of Biosciences, University of Birmingham, United Kingdom, B15 2TT. Dates: 7th – 9th October 2024 Event type: On-site, hands-on Early bird rates: Ending 27th Sept 2024. Bursaries available for a limited number of eligible participants (courtesy of the Centre for Environmental Research and Justice (CERJ), University of Birmingham). This is a new and specialised course in the BMTC portfolio, aimed at PhD, Post-Doctorate and Early Career Researchers pursuing toxicological studies with metabolomics. Through a combination of lectures, hands-on laboratory sessions and data analysis workshops (underpinned by a toxicological case study), participants will: ⏺ Be introduced to the exciting field of metabolomics and learn how it can empower 21st century toxicological research, e.g. - quantifying internal concentrations of chemicals in exposed biosystems - exploring biotransformation products of exposure chemicals - generating hypotheses relating to an exposure compound’s molecular mechanism(s)/mode(s) of action ⏺ Explore experimental design principles that affect successful execution of metabolomics-based toxicology research and relevance of associated results. ⏺ Learn the fundamentals of sample collection and extraction, gaining greater understanding of their importance in metabolomics data quality and content. ⏺ Gain hands-on experience in setting up, calibrating and operating Thermo Scientific's Q Exactive Plus and Orbitrap Tribrid ID-X mass spectrometers, coupled with Vanquish liquid chromatography systems, for: - full-scan LC-HRMS profiling of biological extracts - targeted analysis and quantification of exposure compounds - tandem and multi-stage mass spectrometry analyses for annotation/identification of exposure compounds, biotransformation products and endogenous metabolites. ⏺ Learn how to process and analyse metabolomics data, with a focus on toxicological applications and the associated challenges therein. I’m excited to share that I'll be presenting my recent research in an upcoming webinar hosted by the Society of Toxicology's Computational Toxicology Specialty Section (CTSS). This opportunity follows my receipt of the CTSS Elsevier Student Award at the SOT Annual Meeting in Salt Lake City earlier this year—a recognition for which I’m incredibly grateful. Along with fellow award recipients, I will present my work tomorrow, November 14th, at 11:00 AM EST. My presentation is titled: "Predicting Chemical Immunotoxicity Through Data-Driven QSAR Modeling of AhR Agonism and Related Toxicity Mechanisms."
