Once more into the IgA fray. We just learned Jade Bio and Aerovate will merge to focus on developing a potential “best-in-class” treatment for IgA nephropathy. Jade is the fourth spinout of Paragon Therapeutics, Inc. to go public via a reverse merger and the third this year, following Oruka Therapeutics and Crescent Biopharma. Paragon specializes in discovering promising new antibody drugs, which it then licenses to newly created companies to advance their development. In Jade’s case, JADE-001 targets a protein known as APRIL that’s thought to play a major role in the development of IgA nephropathy, which over time erodes kidney function. Jade aims to dose its drug every two months, compared to anti-APRIL antibodies being developed by Otsuka Pharmaceutical Companies (U.S.) and Novartis that are dosed once per month or once every two weeks. (Otsuka last week said its antibody, sibeprenlimab, succeeded in a Phase 3 trial, while Novartis expects data for its drug, zigakibart, in 2026.) The combined company will be named Jade Biosciences and will be led by Tom Frohlich, Jade’s current Chief Executive Officer (CEO). Jade’s existing Board of Directors will remain in place, chaired by Eric Dobmeier, former President and CEO of Chinook Therapeutics and current Venture Partner at Samsara BioCapital. Board members include Lawrence Klein, Ph.D., CEO of Oruka Therapeutics; Erin Lavelle, former Chief Operating Officer and Chief Financial Officer at ProfoundBio and Eliem Therapeutics; Chris Cain, Ph.D., Director of Research at Fairmount and Tomas Kiselak, Managing Member at Fairmount. Tryptamine Therapeutics is pleased to announce the completion of its Phase 2a clinical study for the treatment of Fibromyalgia. The study was initiated by the University of Michigan (‘UOM’) in January 2024, one of the top-ranked public universities in the USA, in collaboration with Tryptamine Therapeutics Fibromyalgia represents a large global market opportunity affecting over 1m people in Australia and an estimated 10 million people in the USA. Researchers from UOM plan to present the clinical trial results at an upcoming global meeting (International Association for the Study of Pain Conference) to be held 5 to 9 August 2024 in the Netherlands. It is expected that this will provide Tryp Therapeutics with exceptional exposure to leading industry participants. Results of this study will inform future clinical studies in fibromyalgia patients using TRP-8803 (IV-infused psilocin), Tryp’s proprietary psilocin formulation which may address many of the limitations associated with orally administered psilocybin. TRP-8803’s potential advantages include a significant reduction in onset of the psychedelic state, more precise control of the depth and duration of the psychedelic experience and a reduction in the overall duration of the intervention to a commercially feasible timeframe. Additional clinical trials continue to progress well with the first 3 participants having completed dosing in TRP-8803’s healthy human volunteer study, while work towards commencement of the exciting Phase 2a trial with Harvard University's Massachusetts General Hospital (MGH) for treatment of irritable bowel syndrome continues to advance. Private Market Financings Week Ending 8/8/2024 💰 Waypoint Bio received $14.5 million in Seed funding. The company is focused on developing cell therapies for solid tumors leveraging their proprietary in vivo spatial pooled screening technology. The round was led by Hummingbird Ventures with participation from Recode Ventures and Fifty Years 💰 Granza Bio (YC W24) bio successfully raised $7 million in Seed funding to advance the development of their therapeutic delivery platform. The new capital will enable Granza Bio to advance its research and develop its therapeutic cargo delivery system, potentially leading to new drug therapies. The round was led by Felicis and Refactor with participation from Y Combinator, Metaplanet, Pioneer Fund, and others. 💰 SciRhom, a German antibody company, raised a $70 million Series A with participation from Andera Partners, Kurma Partners, Hadean Ventures, MIG Capital and Wellington Partners. SciRohm will look to advance its lead monoclonal antibody program through a first-in-human trial set to start in the second half of the year. The Phase 1 trial will study SciRhom’s lead candidate, SR-878, in healthy volunteers in Austria later this year. 💰 Myricx Bio, a London based biotech, raised a $114 million Series A with participation from Novo Holdings and Abingworth. Myricx made a strategic change away from its IV and small molecule approach and is opting instead to attach its payload to antibody-drug conjugates. 💰 Circle Pharma, Inc., a Bay Area biotech developing macrocycles for "undruggable" targets, is seeking approximately $117 million for a Series D. The company has already secured about $54 million from a dozen investors. Previously, Circle Pharma raised $66 million in a Series C round in June 2021. Investors in the company include The Column Group, Nextech, Pandect Bioventures, Eli Lilly and Company, and Pfizer. 💰 Beacon Therapeutics has raised $170 million to potentially bring a rare ophthalmology gene therapy to market in competition with Johnson & Johnson. Participants in Beacon's recent financings include existing funders Oxford Science Enterprises, Forbion, and new investors Advent Life Sciences and TCGX. 💰 Cartesian Therapeutics, a Maryland-based mRNA cell therapy biotech, advanced a subsequent Phase 2 trial while raising $130 million in a PIPE from HBM Healthcare Investments AG, The Invus Group, Schooner Capital, and Surveyor Capital. The company’s lead investigational medicine helped patients with an autoimmune condition that has caught the attention of many biopharmas in recent years. 📑A recent May 2024 paper by Michel Sadelain’s group published in Cell Press Molecular Therapy features the over expression of GLUT1 as a way of improving CAR-T potency, persistence, and reducing exhaustion! 💪🏽😮‍💨 How!? GLUT1 achieves this through promoting T stem cell-like memory phenotypes and enriching for genes involved in glycolysis, mitochondrial respiration, and other memory precursor phenotypes, which the group shows in animal models 🐁 has a direct effect on improving CAR-T performance. This work was built off of research by Sadelain’s group presented at American Society of Hematology and published in Blood 🩸 in 2023 showing overexpression of GLUT1 enhances T-cell expansion and therapeutic efficacy. 🌟🧑🏻‍🔬Our team has demonstrated with a number of partners a direct relationship between GLUT1 over expression induced by a hypoxia and hyperbaric environment and some dramatic improvements in T-cell expansion rates and therapeutic potency. Manufacturing doesn’t have to be the problem. Under the right conditions, it offers the solution to less exhausted, more potent cells, in less time.
