Gupta, accompanied by her ministers, state BJP in-charge Baijayant Panda, and state president Virendra Sachdeva, took charge at the Delhi Secretariat following a grand oath-taking ceremony at Ramlila Maidan.
"I assure that not a single day will be wasted in realising our mission of a developed Delhi," Gupta told reporters.
Earlier, the first-time MLA from Shalimar Bagh was sworn in as chief minister by Lieutenant Governor V K Saxena, with her council of ministers also taking their oaths.
The ceremony was attended by Prime Minister Narendra Modi and other top BJP leaders, but no AAP leaders, including former CM Arvind Kejriwal, were present. In the evening, Gupta performed the 'Yamuna Aarti' at Vasudev Ghat with her cabinet.

Both parties targeted AAP convener and former CM Arvind Kejriwal for the severely polluted Yamuna.
Rekha, along with Delhi BJP state president Virendra Sachdeva and her cabinet ministers – Parvesh Sahib Singh, Ashish Sood, Manjinder Singh Sirsa, Ravinder Indraj Singh, Kapil Mishra, and Pankaj Kumar Singh – performed the 'Maa Yamuna Puja' before conducting the Aarti, just before the first cabinet meeting. Ram bhajans echoed at the ghat, with chants of "Jai Shree Ram" and "Yamuna Maiya Ki Jai."
Earlier, Rekha, the first-time MLA from Shalimar Bagh, took her oath as CM in a grand ceremony at Ramlila Maidan, administered by Lieutenant Governor V K Saxena.
