🚨 AstraZeneca and Daiichi Sankyo US announced positive topline results from the Phase III DESTINY-Lung04 trial. 💊 Enhertu (trastuzumab deruxtecan) demonstrated a statistically significant and clinically meaningful improvement in progression-free survival as first-line treatment for patients with unresectable, locally advanced or metastatic HER2-mutant non-squamous NSCLC, compared with platinum-pemetrexed chemotherapy plus pembrolizumab. The safety profile was generally consistent with the known profile of Enhertu, with no new safety concerns identified. AstraZeneca is discontinuing the Phase III eVOLVE-Lung02 trial evaluating volrustomig plus chemotherapy versus pembrolizumab plus chemotherapy as first-line treatment for metastatic NSCLC with PD-L1 <50%. Following a planned review, the Independent Data Monitoring Committee concluded that the volrustomig combination was unlikely to meet the dual primary endpoints of progression-free survival and overall survival in patients with PD-L1-negative tumors (<1%). AstraZeneca reported no new safety signals. Other Phase III studies of volrustomig in cervical cancer, head and neck squamous cell carcinoma, and mesothelioma will continue as planned. 💊 AstraZeneca and HUTCHMED have reported positive results from the Phase III SAFFRON trial (NCT05261399) evaluating Tagrisso (osimertinib) plus Orpathys (savolitinib) in patients with EGFR-mutated, locally advanced or metastatic NSCLC with high MET overexpression or amplification following progression on Tagrisso. According to AstraZeneca, SAFFRON is the first global Phase III trial to show significant PFS and OS benefits in this setting. The safety profile was consistent with the known profiles of the individual treatments, with no new safety findings reported. Detailed efficacy and safety results have not yet been disclosed and are expected to be presented at a forthcoming medical meeting.
