To test whether Synteins™ behave differently than natural proteins, the Abiologics team compared sequence-matched L-proteins and Synteins™ in protease-rich environments where proteins are rapidly degraded. L-proteins were degraded within hours in rodent serum and became undetectable after just two minutes in simulated intestinal fluid. Synteins™, by contrast, remained intact for at least seven days in serum and five days in intestinal fluid. These findings demonstrate a fundamental property of D-proteins: intrinsic resistance to protease degradation. That durability could expand the range of therapeutic applications and delivery routes that may be possible, including oral administration. To study whether engineered circular RNAs could move systemically, Terrana Biosciences’ team applied an engineered circular RNA construct, visualized using a yellow reporter signal, to the stem of an eggplant leaf and tracked its signal over time. Four weeks after localized delivery, the RNA was moving through the leaf vasculature and appearing in distal tissue, evidenced by significant yellowing of the upper leaves.
