Red Riding Hood is training in the group of Sister Hoods, when she and the Wolf are called to examine the sudden mysterious disappearance of Hansel and Gretel.

Hood vs. Evil will find a teen such as Red Riding Hood who will be training in a distant land with a mysterious, covert group called the Sister Hoods. In the world of fairy tales and stories, a team of characters must learn to work together as a team to investigate the disappearance of a couple of missing people. Secrets from the past are revealed which could change everything and everyone who lives in the area.—RECB3

Red, Wolf, Granny and Squirrel Twitchy work for Nicky Flippers (David Ogden Stiers) at the Happily Ever After secret agency, dedicated to making happy ending for everyone. Together they defeated Boingo Bunny. Red is away for training with the Sisters of the Hood and hence the team is in a bit of disarray.

Wolf (Patrick Warburton), Granny (Glenn Close), and Twitchy (Cory Edwards) are on a rescue mission to save Hansel (Bill Hader) and Gretel (Amy Poehler) from an evil witch (Joan Cusack) who has kidnapped the 2 kids and kept them hostage in a ginger house. Wolf is not happy that Red has been sent for training & is no longer with the team.

The plan goes wrong when Wolf & Twitchy ignore orders & attack the house on their own. Granny risks her own life to save them but is kidnapped by the witch in the process who flies away with the kids. Granny tries to pursue the witch on her bike, and manages to catapult herself to the witch's broom, which had jet engines attached to it. The witch handcuffs Granny and flies away despite efforts from Wolf and Twitchy to save her. Granny tells Wolf to look for Red and that only she would know what to do.

Meanwhile, Red (Hayden Panettiere) is in training with a mysterious group called the Sisters of the Hood. Red is trained in various martial arts techniques, including Kung Fu which she uses to defeat a variety of opponents in training (including a troll named Moss (David Alan Grier)). Red still gets riled up when she is called a helpless little girl. Due to her anger Red fails to complete her combat assignments despite being the superior fighter (the assignment was to deliver the cupcakes intact, while defeating the troll, but in her anger, Red smashed the cupcakes). Nicky calls Red and informs her of Granny's abduction.

Red learns that a secret all powerful truffle recipe of the Hood has been stolen from the heavily guarded vault of the Hood. The recipe had been in the Hood for 1000 years and only the best of the Sisters could understand it and recreate the dish called the Norwegian Black Forest Feather Cake Truffle Divine or the Super Truffle. Her Granny/Abigail Puckett was one of the very few Sisters who successfully made the truffle dish from the recipe. The truffle dish gives its owner unlimited powers in strength and thought and speed.

However, there is a secret ingredient that is never written down & is not told to Red. Iana (Debra Wilson) the head of the Hood tells Red that when you are a true sister of the Hood, you automatically know the secret ingredient and speculates that this is why Granny has been abducted (as she knows the secret last ingredient).

Red teams up with Wolf and Twitchy to find the recipe and save her grandma, but she and Wolf can't get along and the team splits up. Meanwhile the witch, Verushka (Joan Cusack), who stole the truffle recipe from the Hood vault, tries to force Granny into making the truffle dish for her. Granny resists but Verushka threatens Hansel & Gretel's death, hence Granny relents.

Red & Wolf follow the trail of The Giant (Brad Garrett) who seems to be ordering the ingredients on the truffle recipe. Nicky asks Red and Wolf to be partners, overriding their objections to working with each other. They visit the Giant's bar called beanstalk & talk to his harp named Jimmy 10-Strings (Wayne Newton) (who used to be Granny's informer). The Giant corners Red and Wolf before they can talk to the Wolf, but Twitchy takes advantage of the distraction and carries harp out of the bar. While escaping from the bar, the Harp tells them that the Bunny Boingo gives the order to the Giant & he delivers. Eventually, the Giant pursues the trio and catches up to them, but Red asks everyone to jump from the beanstalk to escape. Red saves everyone by fashioning a parachute out of her cape. Boingo is in the London Bridge Asylum and Prison, so Red & Wolf visit him & find that Verushka passes her order to Boingo written inside a book, that she herself delivers under her guise as a cleaning lady. Red and Wolf continue to blame each other for their mistakes and failures along the way. Wolf leaves Red citing that he works better alone anyways.

Granny escapes by distracting the witch. She asks for a titanium mixing bowl and then criticizes the quality of the equipment and the ingredient. The witch has no option but to agree and goes off to hunt for the ingredients. Granny uses this window of opportunity to escape her holding cell. Granny finds Hansel and Gretel on the 1093rd floor of the witch's castle but learns that they are actually the masterminds behind the plot. They want the truffle dish to become unstoppable & to wreck their vengeance on the world. Granny is recaptured and learns that Verushka was an old classmate of hers when they were both in training with the Sisters of the Hood; Verushka was always second behind Granny's accomplishments. This is why Verushka stole the recipe right after her graduation and spent the last 60 years trying to learn the secret ingredient.

Red discovers via tracing a piece of clothing she took after she visited Boingo in jail. The cloth had a tag that said DCT and she tries to ask everyone in the city what it meant. A weasel beggar tells Red that DCT stands for Dark Castle Towers and takes her to the location.

Red calls Nicky who promises to send an entire backup squad to help Red. This call is intercepted by Hansel and Gretel, and they are ready. Hansel & Gretel initiate their plan to eliminate all their enemies (they attack the HEA HQ) including Wolf & Twitchy (Wolf was with Twitchy at his trailer in the woods). The evil pig agents arrive at the trailer park but fire their grenade launcher at a different trailer. The pigs realize their mistake and pursue the trailer to shoot it out of the sky, but they do survive by jumping out of the trailer just before the grenade hit. Red is captured when trying to infiltrate Verushka HQ.

Wolf & Twitchy reached the HEA HQ to find Nicky who tells them Red's location. Nicky says that all of HEA's assets are out of commission and Wolf and Twitchy are the only ones left. Red arrives to save Red & together they reach Verushka's HQ with the help of their strong friend Klaus (Cameron Barnes) (who brings his gang back together including Helmut (Mike Disa) and Heidi (Heidi Klum)). Red enters an empty room with the incomplete truffle dough & tastes it & accidentally reveals the final ingredient for the truffle recipe (macadamia nuts) at which point Hansel, Gretel & Verushka reveal themselves & their plans for the super truffle.

Hansel & Gretel make several Super Truffles & plan to sell them to many villains to become rich. Hansel and Gretel eat the truffles transforming into giants, so that they cannot be stopped by anyone and go on a rampage through the city. Hansel and Gretel betray Verushka and Granny convinces her to join forces with her, Red, Wolf, and Twitchy.

They trick Hansel and Gretel into eating more truffles which make them so obese that they can no longer move their arms and legs. Hansel and Gretel are then arrested and condemned to eat broccoli and get a lot of exercise, especially on a treadmill. Afterwards, Nicky recruits the team for another mission about a man with a flute being chased by 10,000 mice.
