Previously, we highlighted the need to bridge discovery and manufacturing in multispecific antibody (MsAb) development. Given their structural complexity, MsAbs are more challenging to manufacture than traditional monoclonal antibodies. Therefore, early assessment of manufacturability at the discovery stage is imperative to reduce the risks associated with process scale-up at later stages. At #BTI, Associate Professor Ng Say Kong and his team have established a multi-parameter approach to address the challenge. An automated parallel bioreactor system is utilised to systematically evaluate large numbers of clones across diverse process conditions within a tightly controlled environment. This accelerates the identification of antibody candidates which demonstrate robust performance and strong scalability potential. A proprietary high‑throughput media component screening platform sheds light on how individual media components can influence cellular phenotypes. Close collaborations with A*STAR BTI's Analytical Science & Technology research groups enable the team to incorporate multi-omics analysis into their media optimisation work. This end-to-end approach supports an efficient and systematic screening of potential MsAb candidates with strong scalability across various media and process conditions. This streamlines the development timelines and improves the likelihood of success for MsAb development. Multispecific antibodies are opening a new chapter in biologics — but with that promise comes significant complexity. From bispecifics and trispecifics to emerging next-generation formats, multispecific antibodies are engineered to engage two or more targets or epitopes simultaneously. This opens up a range of therapeutic possibilities: 1️⃣ redirecting T cells to tumours 2️⃣ co-blocking immune pathways 3️⃣ shuttling across biological barriers while engaging a disease target. Clinical validation of this modality continues to grow, with approved therapies such as blinatumomab, faricimab, and mosunetuzumab, as well as hundreds of bispecific candidates in development. However, the real challenge extends beyond the mechanism of action. A promising multispecific antibody must also become a stable, developable and manufacturable biologic. That means addressing key challenges in: ▪️Molecular design and correct chain pairing ▪️Biophysical liabilities such as aggregation and stability ▪️Purification and process scalability ▪️Advanced analytical characterisation ▪️Integration between discovery, development and manufacturing At #BTI, we are strengthening our capabilities across antibody engineering, developability assessment, product characterisation, process development, and predictive hybrid modelling. Emphysema happens when there is insufficient alpha-1-antitrypsin (A1AT) to neutralise neutrophil elastase, a component that is essential in fighting infections in the lung. Current treatment for emphysema involves administering A1AT derived from plasma at high frequency and dosage amounts. However, this treatment is costly due to the limited supply of human plasma from donors. Wenqin Tang, Dr Ng Say Kong and team at #BTI have found a solution to scale up the production of recombinant human A1AT (rhA1AT). This can potentially reduce the reliance on human plasma derived A1AT. CHO single cell clones producing A1AT were evaluated for their production stability, in terms of specific productivity and growth rate. The highest performing clone was selected for production in both shake flask and a 10L single-use orbital shaking bioreactor. The growth profiles were observed to be similar at both scales, thereby demonstrating the scalability of the production process. The harvested rhA1AT was purified to remove protein aggregates and fragments, resulting in an increased activity level from 37% to 65%. Finally, the identity of the purified rhA1AT was verified via peptide mapping. Scaling up rhA1AT production could provide a more consistent supply for patients that is both cost-effective and safe. In addition, utilising single-use bioreactors for scalable rhA1AT production allows production to take place in a multi-product facility, thereby reducing initial capital investment risks for manufacturers.
