Beyond the 28% ORR Ceiling: Why CRC Needs a New Generation of ADCs. The Colorectal Cancer (CRC) landscape is evolving rapidly. With one ADC against cMET offering great promise for 25% of the patients, and several others targeting CEACAM5, CDH17, and EpCAM, the industry is making strides. However, looking at the data, a clear challenge remains. While these 3 targets cover a large patient population on paper, clinical results disclosed so far have largely plateaued, remaining below a 28% ORR. For patients with CRC cancers, there is still significant unmet therapeutic potential. The data is out: our first-in-class ADC, ATL-024, will be featured at the upcoming AACR Annual Meeting in San Diego! Following our recent launch, we are excited to share the scientific foundation of Antelope Therapeutics. The public abstract for our poster presentation is now available (link in the comment), highlighting the preclinical performance of ATL-024 in gastrointestinal cancers. Key takeaways from our findings include: 🔹 Best-in-disease potential: ATL-024 demonstrated a compelling efficacy in multiple models, including in a 16 CRC models SMT-PDX study. The tolerability in NHP was also outstanding with the highest HNSTD ever disclosed for CRC ADCs. 🔹 Broad GI applicability: Robust efficacy was observed not only in CRC but also in Pancreatic and Gastric cancer models.
